It's real cars or CGI?

Both, I think. Basically, it looks like whenever the cars are stationary (or moving in a straight line, I suppose), they're real. But every other time, they're CGI (the movie was flimed entirely with a green screen).

When is this film set? Almost all cars are from the 1950's, except for the 80's Ferrari and Mercedes.

It's meant to be in a '90s era, when most (or nearly all) of the stories were written. Sin City is simply home to a large number of classic cars, presumably because of its desert location with infrequent rain showers and rare winters.

In the director's cut, there's at least three other Bel Airs, and a Cadillac ambulance that was used to take Hartigan to the hospital.

I'd say, based on the single headlamps, D-pillar and window, windshield, hood, grille and tailfins, it's a 1956 or 1957 Miller-Meteor Futura Ambulance.

Should the ambulance be added? It could be classified as Seen only in Deleted Scene (although it is seen for several seconds and transports a main character to the hospital).

The trailer for the sequel has just come out and there are some pretty awesome cars in it https://www.youtube.com/watch?v=RIFqHn_Ul0M I just really hope that the Tucker Torpedo from the origenal A Dame To Kill For graphic novel is going to be in the movie

It looks like it will. I've found an interview with the guys from Ida Automotive of New Jersey that states they built two replica Tuckers for the movie.
Also, there is a teaser video on youtube that shows some of the work on the car... but sadly nothing of the finished car.

There were few bits in colour; the taillights on some of the cars, the police cars' flashing roof light, Goldie's blonde hair, the blanket on Marv's love heart shaped bed, the Customer's dress and eyes, Becky's eyes, the police motorcycle's flashing lights, the beam of light from the motorcycle cop's torch, the neon signs on the buildings, the blood from Marv's wounds, the rose tattoo on the face of one of Kevin's victims, Dwight's Cadillac, Jackie Boy's Imperial, Roark Junior's alter ego, his alter ego's blood and the cars' brake lights.

Some were, there were other objects in different colours:

. Detective Jack "Jackie Boy" Rafferty's Imperial was a light blue.

. Yellow Bastard's Cadillac was black.

. Kadie's Saloon neon sign had blue and yellow.
